Resistance is a cardi nal feature of immunity\, whereby soluble and cellular components of the i mmune system sense and target pathogenic microorganisms for containment\, destruction\, or expulsion. In addition\, multicellular organisms deploy a nother defense strategy\, referred to as disease tolerance\, which does no t target microbes directly. This evolutionarily conserved strategy relies on stress responses that rewire host energy metabolism. At the Inflammatio n Laboratory\, researchers aim to identify and characterize how these stre ss responses engage inter-organ communication processes that rewire energy metabolism in order to limit the fitness costs associated with infections caused by different classes of microbes. The team applies these techniques to vari ous types of biological data\, including protein and DNA sequences\, data from high-throughput genomic assays such as ChIP-seq and Hi-C\, and tandem mass spectrometry. Prof. Noble’s research group is currently developing methods for analyzing shotgun proteomics data\, for characterizing protei n function\, structure and interactions\, and for understanding the struct ure and regulatory influence of chromatin.\n\nWilliam Stafford Noble is a Professor in the Department of Genome Sciences and in the Paul G. Allen Sc hool of Computer Science and Engineering at the University of Washington. He received the Ph.D. in computer science and cognitive science from Unive rsity of California\, San Diego in 1998. He is an editor of the Statistical Modelling Journal and a member of the Clinical Ethics Committee in Marburg.\n\nLarge-scale cohort studies are a key resource for understanding how genetic predisposition interacts with environmental and lifestyle factors to influence disease risk over time. A t the same time\, incorporating high-dimensional genomic data into such st udies remains challenging\, particularly when aiming to move beyond standa rd polygenic risk score (PRS) approaches based on GWAS summary statistics. In his talk\, Prof. Mayr will present boosting-based methods (Mayr et al. 2014) for deriving and extending PRS using individual-level data from lar ge-scale cohorts. Building on the snpboost framework (Klinkhammer et al.\, 2023\, 2024)\, he illustrates how statistical boosting enables scalable e stimation of polygenic models while accounting for the complex structure o f genetic data.  A particular focus will be on recent work on boosting di stributional regression for detecting gene–environment interactions (Wu et al.\, 2026). By modeling not only the mean but also the variability of clinical outcomes\, this approach allows the identification of genetic var iants that may modify the effect of environmental exposures\, thereby supp orting more targeted prevention strategies. Her work contri buted to the initiation of a clinical trial (NCT04676477) based on mechani stic studies of HER3-targeted therapy in EGFR-mutated lung cancer. She has a longstanding interest in immune regulation with a particular focus on how CTLA-4 and CD28 operate t o control T cell immunity. Her group aims to understand the pathways that regulate the development of autoimmune diseases\, like type 1 diabetes (T1 D)\, with the goal of developing new immunotherapies. She is particularly interested in the homeostasis and function of regulatory T cells\, and how these cells use CTLA-4 to restrict conventional T cell activation and lim it damaging immune responses. The overarching aim of her group’s researc h is to use mechanistic insights to develop new immunotherapeutic strategi es. Dr Vaahtomeri is also affiliated with the new Centre of Excellenc e in Immune–Endothelial Interfaces (IMMENs). His recent work focuses on immune cell migration\, lymphatic endothelial guidance cues\, and mechani sms controlling leukocyte entry into lymphatic vessels.\n\nCell migration guidance is one of the most complex processes in cell biology. For example \, development and immunity are critically dependent on timed and efficien t arrival of cells to correct locations. In immunity\, leukocyte’s funct ion relies on continuous change of tissue compartments. In this context\, one of the migration events that requires the highest accuracy is the leuk ocyte transmigration across the endothelium. Previous studies have shown t hat\, for example\, neutrophils prefer to transmigrate the blood endotheli al multicellular vertices (i.e. the cell junction where multiple endotheli al cells meet). These junctions are characterized by discontinuities in ce ll-cell junction proteins and increased ICAM expression\, but it is not kn own how leukocytes are guided to these sites. To address this\, we utilize d chemokine CCL21-dependent antigen-presenting dendritic cell (DC) entry i nto the lymphatic system as a model of guided transmigration.  By using i n vivo\, explant and primary cell co-culture assays of cell migration\, to gether with advanced imaging methods\, we have identified lymphatic endoth elial multicellular vertices as sites of leukocyte entry into the lymphati cs. Further\, we show that the entry sites are determined by spatially con fined guidance cue exocytosis\, controlling dendritic cell migration to de rmal lymphatic vessels. Altogether\, our results exemplify how localized c hemokine exocytosis is critical for cell migration guidance cue formation.
